热疗逆转乳腺癌细胞系耐药的实验研究()

摘要:
目的
探讨热疗对体外培养的乳腺癌细胞多药耐药的逆转作用。
方法
对乳腺癌多药耐药细胞系MCF-7/ADR给予42.5℃ 0.5h的加温处理。MTT法检测热疗前后细胞对阿霉素(ADM)、5-FU、环磷酰胺(CTX)的耐药性的变化,计算半抑制浓度(IC50)、耐药倍数;定量PCR测定热疗前后细胞多药耐药基因mdr1的表达;流式细胞术检测细胞表面P-gp的表达。
结果
热疗后细胞的生长及细胞周期没有明显的变化,但是热疗后MCF-7/ADR细胞对多种临床一线化疗药(ADM 、5-FU及CTX)的耐药性均产生下调效应,IC50分别下降21.2%、14.9%、16.6%(P<0.01)。定量PCR未检测到mdr1表达的明显变化,但是流式细胞仪检测却显示P-gp的表达显著降低。
结论
热疗能够对体外培养的乳腺癌细胞系的耐药性产生逆转作用,增强化疗药的敏感性,其机制可能是热疗在翻译水平下调P-gp的表达。热疗的耐药逆转作用可能为临床化疗开辟新的途径。
