摘要:目的 探讨低毫安电化学疗法(ECT)对乳腺癌MCF-7细胞内钙离子浓度改变与诱导细胞凋亡、坏死及抑制细胞增殖关系。方法 用不同电量处理MCF-7细胞培养6小时和24小时后通过MTT法、GENMED细胞钙离子浓度比色法、流式细胞仪和激光共聚焦分别检测细胞抑制率、细胞Ca2+浓度、观察凋亡和坏死情况。结果 MCF-7细胞抑制率、坏死率、细胞Ca2+浓度与对照组相比均呈明显依次增高(P<0.05),细胞凋亡率在3、5、10C(C为电量库仑)明显依次增高,而20C增高不明显 (P<0.05)。24小时比6小时增加明显(P<0.05)。结论 ECT可抑制乳腺癌MCF-7细胞株的生长并诱导其凋亡,细胞内Ca2+浓度升高可能为其机制之一;低剂量ECT主要诱导凋亡,高剂量ECT主要引起细胞坏死。
Abstract:
[Abstract] Objectives: Study on relationship between breast cancer cell line MCF-7 the inhibitory effects and apoptosis inducing by electrochemical therapy and changes of Intracellular Ca2 +.Methods:6 hours and 24 hours ’’’’’’’’’’’’’’’’after treatment, the inhibitory rate of human breast cancer cells growth was detected by the MTT method, the apoptosis and necrosis percentage were treated by Confocal Laser Scanning Microscope and flow cytometer assay. the change of Intracellular Ca2 + were treated by GENMED Calcium Colorimetric Assay Kit.Results: The inhibitory rate and Intracellular Ca2 + are significantly increased after 6h and 24h of ECT tratment compared with the control( P < 0. 05) . The apoptosis percentage of 3C,5 C,10C ((C:coulomb )groups are significantly increased, The apoptosis percentage of 20C is not obvious increased (P<0.05). The change of the results especially obvious at 24h( P < 0. 05). Conclusions: The study shows that the ECT induces the apoptosis and can inhibit the proliferation , the increased content of Intracellular Ca2 + should play an important role to induce the death of ones, low dose of ECT mainly leads to cell apoptosis and high dose of ECT mainly leads to death of the cell.
